大鼠慢性压迫性脊髓损伤后凋亡调控相关基因p53的表达变化及意义

【摘要】 目的:探讨慢性压迫性脊髓损伤(chroniccompressivespinalcordinjury,CCSCI)后脊髓组织中凋亡相关基因p53的表达变化规律及意义。方法:通过制作Wistar大鼠慢性压迫脊髓损伤模型,应用免疫组织化学方法和原位杂交方法,研究压迫组、假手术对照组脊髓组织中p53mR-NA和p53蛋白表达的灰度值差异。结果:p53mRNA在脊髓受压后4h脊髓组织即有表达,1d后增高,7d后仍有表达,14d后无表达,而p53蛋白受压1d后呈弱阳性,之后无明显表达。结论:p53的表达规律提示它参与了CCSCI内源性机制的调控过程。

【Abstract】 AIM:To study the expression and significance of p53 after chronic compressive spinal cord injury(CCSCI). METHODS:On the basis of CCSCI model in Wistar rats,immunohistochemistry and hybridization in situ were applied to detect p53 mRNA and p53 protein in spinal cord tissue in both compression group and sham operation group. RESULTS:p53 mRNA in spinal cord tissue after compression expressed at 4 hours,increased at 1 day and existed at 7 days,but disappeared at 14 days,while p53 protein expressed weakly only at one day and then could not be detected. CONCLUSION:p53 can take part in the regulative process of endogenetic mechanism in CCSCI.
