神经生长因子对内耳缺血再灌注毛细胞损伤的保护作用
Protective effect of nerve growth factor on damage of hair cell induced by inner ear ischemia and reperfusion
【摘要】 目的:探讨神经生长因子(nervegrowthfactor,NGF)对缺血再灌注内耳损伤大鼠耳蜗毛细胞的作用。方法:选用健康SD大鼠单纯随机分5组:正常对照组;假手术组;缺血再灌组;盐水对照组(缺血再灌+生理盐水);NGF组(缺血再灌+神经生长因子)。闭塞双侧椎动脉和颈总动脉建立内耳缺血再灌注模型,于内耳缺血30min再灌注2h,1,3,7d不同时间取耳蜗制备样本扫描电镜观察。结果:缺血再灌组、盐水对照组耳蜗外毛细胞损伤严重,静纤毛散乱脱落、倒伏及粘连,外毛细胞散在或灶状坏死缺失,出现不规则空洞缺损,瘢痕形成。NGF组肌注NGF后静纤毛损伤与外毛细胞坏死明显减轻,静纤毛排列整齐,无严重散乱脱落现象,偶见底回静纤毛轻度粘连,外毛细胞表面结构基本正常,未见明显坏死缺失。结论:NGF对内耳缺血再灌注毛细胞损伤有明显的保护作用,NGF能减轻和改善缺血再灌注引起的听觉神经细胞的损伤。
【Abstract】 AIM:To explore the effect of nerve growth factor(NGF) on cochlear hair cell damage induced by inner ear ischemia and reperfusion. METHODS:Healthy SD rats were randomly divided into five groups:normal control group;sham operated group;ischemia reperfusion group;saline control group(ischemia reperfusion+normal saline);NGF group(ischemia reper fusion+NGF).The animal model was made by occluding bilateral vertebral arteries and bilateral common carotid artery.The hair cells of cochlear were observed by scanning electron microscopy(SEM) after 2 h,1,3,7 d reperfusion following 30 min ischemia. RESULTS:In the ischemia reperfusion group and the saline control group,cochlear outer hair cells(OHCS) were severely damaged,stereocilia of OHCS were lost,adhered and arranged disorderly or lodged apparently.There were necrosis,loss,cavity and scar of OHCS in basilar turn and middle turn of cochlear.In the NGF group,the damage of stereocilia and necrosis of OHCS were reduced significantly,stereocilia lined up in order and no severe disorder exfoliation,there were only a few stereocilia gently adhered to each other,the structure of OHCS was nearly to be normal,and there was no obvious necrosis depletion. CONCLUSION:NGF can protect the hair cell against damage induced by inner ear ischemia and reperfusion,and can also reduce and improve the damage of hearing nerve cell induced by ischemia reperfusion.
