增龄对大鼠性激素分泌及生殖腺轴脂质过氧化的影响
Effect of increasing age on sexhormone sexretion and gonad axis lipid peroxidation in rats
【摘要】 目的:探讨Wistar大鼠性激素水平及下丘脑-垂体-性腺轴组织内过氧化物堆积的增龄性改变,为研究衰老的分子机制提供依据。方法:检测3,6,12,18,21月龄Wistar大鼠下丘脑、垂体、卵巢(雌性)、睾丸(雄性)各组织内丙二醛含量以及雌性大鼠血浆雌二醇、雄性大鼠血浆睾酮水平,并与上一月龄组指标作比较。结果:随月龄增长,雄性大鼠血浆睾酮(nmol/L)及雌性大鼠血浆雌二醇(nmol/L)水平逐渐下降,各组与上一月龄组比均相差显著(睾酮:16.60±1.38~1.80±0.49,t=2.65~6.11,P<0.01~0.05;雌二醇:1.84±0.43~0.31±0.05,t=0.30~9.43,P<0.01~0.05);丙二醛的含量(μmol/g)在大鼠下丘脑(0.114±0.011~0.384±0.062)、垂体(0.142±0.021~0.319±0.042)、性腺组织内(卵巢:0.105±0.021~1.031±0.097;睾丸:0.266±0.017~0.982±0.075)明显增多,除6月龄组与3月龄组比无统计学意义外,其余各组与上一月龄组比均相差显著(t=3.12~16.30,P<0.01~0.05)。结论:增龄对性激素分泌的影响可能与生殖腺轴组织内脂质过氧化物的堆积有关。
【Abstract】 AIM:To explore the changes of sex hormones and piling up of lipoxidation in h ypothalamic-pituitary-ovarian axis(H-P-O-A) with increasing age in Wistar r ats, provide evidence for studying of senescent molecular mechanism. METHODS: Malondialdehyde (MDA) contents in H-P-O-A, and levels of estradiol and testosterone in plasma were detected when Wistar rats were 3,6,12,18 and 21 months old. Then the scores were compared with the pre-months-old rats group . RESULTS: With the increase of month,the levels of estradiol in female and the levels of testosterone in male rats were significantly lower than the pre-month s-old rats group(testosterone:16.60±1.38-1.80±0.49, t=2.65-6.11,P< 0.01-0 .05,estradiol:1.84±0.43-0.31±0.05,t=0.30-9.43,P< 0.01-0.05),while the MDA c ontents(μmol/g)in H(0.114±0.011-0.384±0.062)-P(0.142±0.021-0.319±0.0 42-0.105±0.021-1.031±0.097 and 0.266±0.017-0.982±0.075)-A were markedly higher.All groups were significantly different from pre-months-old group(P< 0. 01-0.05,t=3.12-16.30),except 6-month group and 3-month group because there w as no statistic meanings.
