血小板源生长因子对瘢痕成纤维细胞胞外信号调节激酶的影响
Effect of extracellular signal-regulated kinase in scars fibroblasts by platelet derived growth factor stimulation
【摘要】 目的:分析血小板源生长刺激因子(PDGF)刺激后各种成纤维细胞中胞外信号调节激酶(ERK)的表达及磷酸化情况,研究瘢痕特别是瘢痕疙瘩发病过程中ERK的作用。方法:应用免疫印迹(IB)方法检测5例瘢痕疙瘩成纤维细胞(KFB)、瘢痕疙瘩周边相对正常皮肤成纤维细胞(rNHDF)及正常人皮肤成纤维细胞(NHDF)的ERK表达及其蛋白磷酸化。结果:①无刺激条件下,ERK在KFB,rNHDF及NHDF中的表达及磷酸化无差异。②PDGF-BB刺激后,3组成纤维细胞ERK磷酸化增强,非磷酸化含量无明显改变。③DGF-BB刺激后,KFB中ERK蛋白磷酸化明显高于其他两组成纤维细胞。结论:瘢痕疙瘩细胞外基质的过度沉积可能与KFB内PDGF结合位点自身磷酸化和ERK的磷酸化增强有关。
【Abstract】 AIM:To analyze the expression and phosphorylation of various kinds of extracel lular signal-regulated kinase(ERK) in fibroblast by platelet derived growth fac tor(PDGF) stimulation and to investigate effect of ERK in procedure of episode o f scars especially keloids. METHODS:Studies on phosphorylation were performed using five different keloid fibroblasts (KFB,n=5) and their peri-keloid relatively normal fibroblasts(rNHD F,relatively normal human dermal fibroblast,n=5), and normal human dermal fibr oblast(NHDF,standard control) cell cultures at the protein level by using immun oblotting(IB) analysis. RESULTS:The expression and phosphorylation of ERK in KFB?rNHDF and NHDF had n o differences under the none of stimulation; Phosphorylation of ERK in fibroblas t of three groups increased after stimulation of PDGF-BB, however, the content of non-phosphorylation had no significant difference; Proteins’ phosphorylation level of ERK in KFB were obviously higher than that of other two groups after s timulation of PDGF-BB.
