大鼠烧伤感染模型肺组织热应激蛋白70的表达及意义

【摘要】 目的:了解实验大鼠热应激蛋白70(Heatstressproteins,HSP70)的表达与烧伤脓毒症的关系,探讨其作用机制。方法:将36只雄性Wistar大鼠随机数字表法分成6组,每组6只。其中4组以92℃水,18s烫伤(烫伤面积20%),腹腔注射乳酸林格氏液休克复苏,48h后尾静脉注射绿脓杆菌PA103悬液0.5mL(菌量1.5×109CFU/mL)制成大鼠烧伤感染模型,分别对成模后0,4,24,48h时相点进行检测;另以正常组及烧伤48h组作为对照。以免疫组化法及WesternBlot检测各组肺组织HSP70的表达,同时以偶氮显色法检测各组血浆内毒素水平,并对大鼠肺组织进行大体及镜下观察。结果:Wistar大鼠在尾静脉注射绿脓杆菌PA103悬液后,其血浆内毒素水平迅速上升。与正常对照组相比,烧伤感染组大鼠的血浆内毒素水平明显升高(t=2.2281,P<0.05)。正常大鼠肺组织HSP70表达较弱,表达部位在肺泡上皮及支气管上皮黏膜细胞的胞质。在烧伤感染条件下,大鼠肺组织HSP70表达逐渐增强,至成模后24h,HSP70表达累积量达到最高峰,并呈现较明显的核表达。成模后48h,HSP70表达逐渐减弱,核表达逐渐消失,主要在肺泡上皮及支气管上皮黏膜细胞的胞质表达。结论:HSP70与烧伤感染后的应激反应有密切关系,其表达规律与热应激反应相似。在烧伤感染的早期(24h内),热应激蛋白表达水平与血浆内?

【Abstract】 AIM:To study the relationship between the expressions of heat stress proteins 70(HSP70) induced by post burn infection and burn sepsis and to explore its mechanism. METHODS:Thirty six male Wistar rats were divided randomly into 6 groups, and each group had 6 rats. The model of post burn infection was made in 4 groups according to different hours (0, 4, 24 and 48 h respectively), while the other two groups were the normal group and burned group(48 h) as controls. The rats inflicted with 20% TBSA Ⅲ degree scald(92 ℃ hot water for 18 s) followed by intravenous injection with Pseudomonas aeruginosa(PA103)(1.5× 109 CFU/mL) after 48 h were employed as the model of post burn injection. Immunohistochemical methods and western blot were employed to analyze the expression of HSP70 in rats’ lungs. Concomitantly serum endotoxin levels were determined. And meanwhile the lung’s pathomorphology was observed. RESULTS:After intravenous injection with PA103, serum endotoxin levels of Wistar rats increased rapidly, which were significantly higher than that of the control group (t=2.228 1,P< 0.05). In the normal control group, the expression of HSP70 in rats’ lungs was not intensive, which could be only seen in cytoplasm of pulmonary alveolus epithelium cells and bronchial epithelium mucosa cells.Post burn infection produce increased HSP70 level, and the total content of HSP70 came to a peak level at 24 h after stimulation, while apparent expression of HSP70 in nucleus could be seen. At 48 h after stimulation, the content of HSP70 decreased and its expression in nucleus could not be seen.
