西藏油菜种质资源的RAPD分子标记分析
Analysis of Rapeseed Germplasm Resources in Tibet by RAPD
【摘要】 采用 RAPD分子标记 ,对西藏地区部分白菜型和芥菜型油菜资源的遗传多样性进行了分析 ,结果显示 ,2 2条引物在 10 6份白菜型油菜中共扩增出 2 36条带 ,多态性位点比率为 89% ;2 4条引物在 5 0份芥菜型油菜中共扩增出 2 76条带 ,多态性位点比率为 94 %。通过 U PGMA聚类分析 ,将西藏白菜型油菜分为 和 类群 ;芥菜型油菜分为 、 、 和 类群。品种吉西肥 ,既不属于白菜型油菜 ,又区别于芥菜型油菜 ,它可能属于芸薹属的另一类。
【Abstract】 RAPD markers were employed to analyze the genetic diversity of landraces of Brassica rape and Brassica juncea in Tibet. It was shown that 236 polymorphic bands were amplified by 22 random 10 mer primers in 106 landraces of B.rapa and 276 polymorphic bands in 50 landraces of B.juncea, respectively. Through clustering on RAPD markers by UPGMA (Unweighted Pair Group Method Using an Arithmetic Average), 106 landraces of B.rapa were separated into 2 major groups and 50 landraces of B.juncea were divided into 4 different groups. The variety ’Jixifei’ was very different from either B. rapa or B. juncea. It might belong to other types of Brassica, and need to be further studied. The genetic diversity is very rich in rapeseed landraces from Tibet.
