【摘要】 报道了以猪血为材料制备凝血酶的新方法:先将猪血浆在-20℃条件下冷冻2d,离心除去纤维蛋白原,再等电点沉淀,得凝血酶原,经CaCl₂激活后,通过DEAE—SepHarose FastFlow离子交换柱层析纯化,获得比活为1807.9U/mg的电泳纯凝血酶制剂,其收率为48.9％。该方法生产工艺简单,所得产品纯度高,对凝血酶的生产具有一定的指导价值。更多还原

【Abstract】 A novel method of isolation & purification of thrombin from the porcine plasma has been developed. The process is as following: the plasma was frozen at - 20C for 2 days, isolating fibrinogen through centrifugation , precipitated thrombinogen at pI. Thrombin was purified by DEAE-Sepharose Fast Flow after activating thrombinogen with CaCl2 and electrophoretic pure was obtained. The recovery of the method was 48. 9% with specific activity 1807. 9U/mg. The method has the property of simple process and high purity of the product, which can be wildly used in manufacture.更多还原
