澳洲青苹组培苗生根及移栽技术研究
Studies on Techniques of Rooting and Transplantation of Granny Smith Apple Shoots
【摘要】 以澳洲青苹组培苗为试材,研究了其生根壮苗和移栽技术,结果表明:(1)组培苗上中部茎段生根能力强,是组培苗生根的最佳部位;(2)IBA生根效果明显,IBA、NAA和PP333配合使用效果更佳,在1/2MS附加IBA0.7mg/L、NAA0.1mg/L、PP3331mg/L的培养基上生根且长芽,生根率达79%以上;(3)暗培养能显著促进生根,使程序和工效得到简化和提高;(4)采用了自然光下长苗、生根、炼苗同步进行的快速成苗法;(5)组培苗直接移栽到营养土中减少了移栽缓苗次数,缩短了成苗时间;(6)研制成功两步成苗一次移栽的快繁体系。短期内获组培苗1万多株,培育商品苗2000余株。
【Abstract】 Techniques of rooting and transplantation of apple cultivars Granny Smith shoots were studied. The results showed that: (l)The top of stem of Granny Smith shoots rooted easily,which was priority to root. (2)IBA could induce it root, but if added IBA,NAA and PP333 to the medium, the effect of rooting was better than IBA lonely. 1/2MS+IBA0.7mg/L+NAA0.1mg/L+PP331mg/L was the suitable combination, on which not only buds but also roots could be obtained,and the rate of rooting could be up to 79. 75%. (3)Cultivation in the dark was helpful for the increasing of rooting rate and was critical to root. (4)It was the first time that we created the method of rapid cultivation and rooting in the natural light. The technique was high efficiency, high quality and easy operation. (5)Plantlet was directly transplanted into the nutritional soil which could save the time of cultivation and decline the frequency of transplantation. (6)"Two steps technique" was the successful method to transplantation. Over 10,000 plantlets can be obtained in a short term,and the number of survival was beyond 2 000.
