“硼复方”对过量氟实验大鼠血清脂质过氧化和RNA合成的影响
The effect of “borax compound” on lipid peroxidation and antioxidation in serum and RNA from experimental rats ingested with excess fluoride
【摘要】 目的 探讨“硼复方”对氟骨症的治疗效果及血清脂质过氧化物、抗氧化物质和骨、软骨核糖核酸合成的影响。方法 78只SD大鼠 (4~ 5周龄 ,体重 5 0~ 90 g) ,随机分为对照组、低过剂量氟组、高过剂量氟组及相对应的“硼复方”治疗组。测定过量氟实验大鼠用“硼复方”治疗后血清、四肢骨氟含量 ,血清SOD、ALP、MDA、骨和软骨RNA的变化。结果 ①“硼复方”治疗后高过剂量氟组大鼠血清和四肢骨氟分别降低 4 7.81%和 2 9.15 % ;②过剂量氟组大鼠血清SOD在染氟后显著下降 ,“硼复方”治疗后明显恢复 ;③染氟后高过剂量氟组血清MDA明显升高 ,“硼复方”治疗后降低 ;④治疗前后实验鼠骨与软骨中RNA含量无明显差异。结论 用“硼复方”治疗实验大鼠氟骨症有降低体内氟含量 ,减轻过量氟性的过氧化损伤 ,增强机体抗氧化能力的作用 ,但“硼复方”对过量氟导致的骨和软骨含量的减少无明显改善作用
【Abstract】 Objective To explore the pathogenesis of skeletal fluorosis and the effect of “borax compound” on lipid peroxidation and antioxidation in serum and RNA in bone and cartilage from experimental rats ingested with excess fluoride. Methods A total of 78 Sprague-Dawley (SD) rats were divided randomly into the control (C), low-dose excess fluoride group (LF), and high-dose excess fluoride group (HF). After experimental skeletal fluorosis model of rats was created, LF group and HF group were divided into dose excess fluoride group (LF-HF group) and excess fluoride added “borax compound” group (LFB-HFB group). Fluoride contents in serum and limbs, superoxide dismutase (SOD) activity and malonaldehyde (MDA) contents in serum and RNA contents in bone and cartilage of ribs were assayed. Results ① Compared with that of the rats treated with excess fluoride, the fluorine content in serum and limbs of rats in HFB group was decreased by 47.82% and 29.15%, respectively; ② SOD activity in serum of rats in LF group and HF group was decreased, and restored after “borax compound” was added; ③ MDA contents in serum of rats in HF group increased obviously, and decreased after “borax compound” was added; ④ RNA contents in bone and cartilage of rats in LFB group and HFB group did not increased significantly compared with those of LF group and HF group. Conclusion “Borax compound” can reduce the damage of lipid peroxidation resulting from excess fluoride, and enhance antioxidation in rats, but adding “borax compound” does not increase RNA contents markedly resulting from excess fluoride in bone and cartilage.
