pH梯度-冻融法制备硝苯地平脂质体

【摘要】 目的 优选硝苯地平脂质体的制备工艺 ,并评价其质量。方法 采用pH梯度 冻融法制备硝苯地平脂质体 ,以包封率和稳定性为检测指标。用SephadexG 5 0微型柱分离 紫外分光光度法测包封率 ;并比较了冻融前后的包封率、粒度分布 ;考察了冻融次数对包封率的影响。结果 本实验制备的脂质体包封率达到 96 .6 5 % ,冻融前的平均粒度为0 .0 98μm ,冻融后的平均粒度为 2 .0 μm。经冻融处理后 ,脂质体包封率和稳定性明显提高。结论 优选得到的脂质体制备工艺稳定可行

【Abstract】 Objective To study the preparation of nifedipine liposomes (NP) and evaluate their quality. Methods The liposomes were prepared by a pH steps degree-freeze thawing method, with the entrapment and stability as indices. The entrapment was studied by Sephadex G-50 minicolumn centrifugation UV-Vis spectroscopy method. Entrapment, and degree distribution before and after freezing were compared, the effect of freezing times on entrapment was studied. Results The entrapment and the average size of NP liposomes were 96.65% and 2.0 μm, respectively. After freezing, both the entrapment and stability were increased. Conclusion The selected formulation and preparation of nifedipine liposomes are practicable.
