硫化砷对MR2细胞组织因子表达的影响
The effect of arsenic sulfide on tissue factor expression in MR2 cell line
【摘要】 目的 探讨硫化砷 (雄黄As4S4,雌黄As2 S3 )对MR2细胞 (维甲酸耐药的急性早幼粒细胞白血病细胞株 )促凝活性 (PCA)和组织因子 (TF)表达的影响。方法 用 30 0 μg·L-1As4S4与 2 μmol·L-1As2 S3 分别与MR2细胞共同孵育 6、12、2 4、4 8、72h ,用复钙时间、ELISA法、半定量RT PCR检测未处理及两种硫化砷处理后的MR2细胞的促凝活性、组织因子抗原表达及组织因子mRNA转录的变化 ,同时检测未处理的NB4、HL 6 0、K5 6 2细胞的促凝活性、组织因子抗原表达作为对照。结果 As4S4和As2 S3 均呈时间依赖方式下调MR2细胞的促凝活性、组织因子抗原的表达及其mRNA的转录。As4S4的下调作用比As2 S3 的作用更明显。结论 下调MR2细胞组织因子的表达并降低其促凝活性可能是硫化砷改善急性早幼粒细胞白血病 (APL)患者弥散性血管内凝血 (DIC)早期出血症状的主要机制之一
【Abstract】 Objective To investigate the effect of arsenic sulfide on procoagulant activity (PCA), tissue factor expression and tissue factor mRNA transcription in ATRA resistant acute promyelocytic leukemia (APL) cell line—MR2 cell. Methods MR2 cells were treated with 300μg·L -1 As 4S 4 and 2?μmol·L -1 As 2S 3. PCA of the treated cells was detected by one stage clotting assay. TF antigen was detected by ELISA and TF mRNA by semi quantitive RT PCR. Results Both As 4S 4 and As 2S 3 could down regulate the membrane PCA, TF antigen and TF mRNA transcription of MR2 cells in a time dependent manner. The role of As 4S 4 was stronger that of than As 2S 3. Conclusion Down regulating TF expression and PCA of MR2 cells by arsenic sulfide may be one of the mechanims of its improvement on DIC related hemorrhage in APL patients.
