枯草芽孢杆菌TH2菌株抗菌物质产生的技术研究
STUDY ON FERMENTATION CONDITIONS FOR THE ANTI—FUNGAL—BACTERIUM SUBSTANCE OF THE ANTIBIOTIC—PRODUCING BACTERIUM STRAIN TH2(BACILLUS SUBTILIS)
【摘要】 通过正交试验,结果表明枯草芽孢生防菌TH2高产抗菌物质培养基的最佳配方为豆饼粉1.5%,玉米粉为0.5%,麦麸0.5%,大米粉0.5%,最佳的培养条件为通气量25ml装瓶量、pH为7.5、温度为28±1℃,这样产生的抗菌物质最多。枯草芽孢生防菌株TH2培养过程中,pH从培养初始的7.0,经下降又逐步上升到48小时的7.8。OD650从培养初始的0.56,逐步上升到36小时的1.10,而后逐渐下降,84小时降到0.85。孢子生成从12小时开始,孢子数达(108cfu/ml),而后逐渐增加,48小时达到高峰,孢子数达58.2(108cfu/ml),随后孢子数逐渐下降,抑菌圈值随着发酵时间增加逐渐增大。
【Abstract】 The present paper dealt with selection of fermentation conditions for the growth of the antibiotic—producing bacterium strain TH2 (Bacillus subtilis). The results showed that the optimal cultural media consisting of 1.5% bean cake powder, 0.5% corn flour, 0. 5% peptone, according the fermentation test, on which the optimal conditions for the fermentation of strain TH2 were pH 7.5 and temperature28±1℃. Like this, anti—fungal—bacterium substance is at most. During the fermentation, pH gradually decreased down from 7.0 at the beginning then increased up to 7.8 at 48 hours. OD650 raised up from 0.56 at the beginning to 1.10 at 36 hours then decreased down to 0.85 at 84 hours. The peak value of spore generation was 58.2 (108cfu/ml) at 48 hours. Inhibition zone along with ferment the time gain to gradually enlarge.
