大鼠主动脉拉伤模型血管壁凝血酶活性的观察
Observation of Thrombin Activity in a Rat Model of Aortic Endothelial Injury
【摘要】 目的 :观察大鼠主动脉拉伤模型血管壁凝血酶活性的变化。方法 :2FFogarty导管拉伤大鼠胸主动脉 ,术后不同时期取血管在体外与纤维蛋白原反应 ,以FPA的生成量代表血管组织的凝血酶活性。结果 :拉伤的大鼠胸主动脉术后 2 4h~ 4 8h凝血酶活性明显增高 ,72h后明显下降 ,但于 72h~ 7天仍维持于较高水平。结论 :内皮拉伤的大鼠主动脉模型动脉壁凝血酶活性明显增加 ,血管壁FPA生成可代表凝血酶活性 ,对再狭窄的研究有一定意义
【Abstract】 Objective: To observe thrombin activity in a rat model of aortic endothelial denudation.Method: A rat model of aorta endothelium denudation was established by inserting and withdrawing a 2F Fogarty catheter.The injured rat aorta of different stage were removed,and immediately reacted with fibrinogen ex vivo.Results: The generation of fibrinopeptide A was determined by ELISA,which represents the activity of thrombin.The thrombin activity of injured arterial wall significantly increased 24~48 h after injury,and maintained until 7 days.Conclusion: Thrombin activity of rat aortic wall increased followed by endothelial injury,which peaked after 24~48 h.This method can be recommended as means in the study of the mechanism of restenosis.
