实验性急性肝损伤血清IL-18、NO的检测意义及作用机制研究

【摘要】 目的 本实验以ConcanavalinA(ConA)诱发小鼠急性肝损伤模型,检测白细胞介素-18(Interleukin-18,IL-18)及一氧化氮(Nitric oxide,NO)血清浓度变化。并探讨此模型中IL-18和NO的关系、意义及作用机制。方法 用ABC-ELISA法及硝酸还原酶法分别检测了20只正常小鼠(对照组)及20只(实验组)小鼠6h血清中IL-18及NO的浓度。结果 实验组小鼠血中IL-18及NO较对照组均明显增高,差异有显著性。经直线相关分析,IL-18与NO呈显著相关性。结论 IL-18在肝损伤中起关键作用,NO对肝损伤亦有重要作用,NO浓度随IL-18表达水平的变化而变化,两者对肝损伤既有独立又有协同作用。IL-18的浓度高低可用于判断肝损伤的严重程度及预后,IL-18使血清中NO水平升高是其致肝细胞损伤的机理之一。

【Abstract】 Objective To detect the concentration of IL-18 and NO in serum of mice with experimental acute hepatic injury induced by ConA and determined the correlation,significance and pathogenesis between IL-18 and NO.Methods IL-18 and NO were detected in 20 normal control mice injected with saline(0.3 ml/mouse)and 20 treatment mice injected with ConA (20 mg/kg/mouse) by ABC-ELISA and nitrose enzyme method. Results The ConA group mice showed significantly higher levels of serum IL-18 and NO than that in normol control group,and there was a correlation between IL-18 and NO. Conclusions IL-18 plays a key role and NO an important role in acute hepatic injury .NO changes in serum due to that of IL-18 and they have both independent effect and synergetic effect on hepatic injury. The severity and clinic prognosis can be judged by the levels of IL-18 in serum. IL-18 makes serum NO increase is one of pathogenesis which make hepatocyte injury.
