【摘要】 目的　观察白血病抑制因子、干细胞因子、碱性成纤维细胞生长因子对人类胚胎干细胞分离克隆的影响。方法　在基础细胞培养液中加入不同的细胞因子组合 ,培养后观察人类胚胎干细胞集落形成数和各组的最高传代数。结果　细胞因子添加与否对人类胚胎干细胞原代培养无显著的影响 ,而对后继培养有显著影响。结论　在人类胚胎干细胞初代培养时可以不添加细胞因子 ,细胞的克隆过程中添加细胞因子有利于细胞的生长。更多还原

【Abstract】 Objective: To investigate the effects of the cytokines on isolation and cloning of human embryonic stem-like cells.Methods: The different combination of cytokines were mixed with the culture medium of cells .After a period, the cloning number of human embryonic stem-like cells and the topmost passage were observed and registered. Results: There was not obvious effect on the primary culture of human ESC but the effect was distinct on the later passage if cytokines were used. Conclusion: In the primary passage , human ESC may survive without cytokines, and the impact of the existence of cytokines is better on the cloning of human ESC than that of its nonexistence.更多还原
