银杏叶提取物对缺血再灌注致鼠脑损伤的脑保护作用研究

【摘要】 目的 探讨银杏叶提取物对鼠脑梗塞面积、神经功能缺失征象及鼠脑缺血边缘区神经细胞凋亡的影响。方法 采用TTC染色、临床神经功能评分、流式细胞分析和原位末端标记等方法 ,对缺血、缺血再灌注及银杏叶提取物治疗组进行比较研究。结果 银杏叶提取物治疗组于相同时间点鼠脑梗塞面积较缺血再灌组明显减小 ,仅表现轻度的神经功能缺损 ,且神经细胞的凋亡较缺血再灌组显著减少 (P <0 0 1)。结论 血小板活化因子受体拮抗剂银杏叶提取物能明显减小鼠脑缺血后脑梗塞面积 ,减轻脑缺血后神经功能缺损 ,显著减少缺血边缘区神经细胞凋亡数量 ,减缓缺血区神经元损害 ,具有显著的脑保护作用

【Abstract】 Objective To investigate the effect of ginkgo biloba extract on cerebral infarct volume, neurologic function deficits and neural cells apoptosis in cerebral ischemic region following focal cerebral ischemia/reperfusion in rats. Methods The ischemia, ischemia/reperfusion and ginkgo biloba extract treated groups were observed by TTC dyeing, neurologic function score, flow cytometry and TUNEL techniques. Results A significant reduction in infarct volume was found in the ginkgo biloba extract treated group as compared with the control group. Furthermore, ginkgo biloba extract could induce a significant amelioration of neural cell apoptosis and neurologic dysfunction following focal cerebral ischemia and reperfusion in rats.Conclusion Ginkgo biloba extract could decrease cerebral ischemic volume and the number of apoptotic neural cells in ischemic region and attenuate neurologic function deficits after ischemia/reperfusion, suggesting that ginkgo biloba extract could protect the brain after cerebral ischemia and reperfusion.
