【摘要】 目的 对抗 CD71人 -鼠嵌合抗体体外抗肿瘤效应进行研究。方法 以表达 CD71分子的 3种人类肿瘤细胞( K5 62 ,CEM和 SMMC- 772 1)为靶细胞 ,以正常人外周血单个核细胞 ( PBMC)为效应细胞 ,在效 /靶细胞比为 5 0∶1的条件下 ,测定嵌合抗体依赖性细胞介导的细胞毒效应 ( ADCC) ;以 CEM和 SMMC- 772 1为靶细胞 ,在有新鲜补体存在下 ,测定嵌合抗体的补体依赖性细胞毒效应 ( CDC)。结果 抗 CD71人 -鼠嵌合抗体 ( D2 C)能够通过识别 CD71分子 ,并通过人抗体 Fc段介导 ADCC。在有新鲜补体存在下 ,介导 CDC。结论 抗 CD71人 -鼠嵌合抗体对 CD71+肿瘤细胞可介导 ADCC和 CDC。

【Abstract】 Objective To investigate the in vitro antitumor effects of anti-CD71 mouse/human chimeric antibody(D2C Ab). Methods CD71 positive target cells (K562, CEM and SMMC-7721) and the effecter cells, freshly isolated normal human peripheral blood mononuclear cells (PBMC), with the target to effecter ratio being 1∶50, were incubated in various dilutions of D2C Ab. Antibody-dependent cell-mediated cytotoxicity test (ADCC) was determined using LDH release assay. Instead of effector cells, complement was added to the target cells (CEM, SMMC-7721) with various dilutions of D2C Ab A method of counting rate for death cell was used for measurement of complement dependent cytotoxicity assay (CDC). Results A significant ADCC was observed with the increased concentration of the D2C Ab. A cytolysis of CD71-positive target cells by the D2C Ab was found in the presence of normal rabbit serum.Conclusion CD71 human/mouse chimeric antibody can mediate ADCC and CDC of CD71 + tumor cells in vitro.
