外源性TGF-β1对兔关节软骨细胞分泌金属蛋白酶的影响
Effects of Exogenous TGF-β1 on Excretion ofMatrix Metalloproteinase of Rabbit Joint Chondrocyte
【摘要】 目的 :探讨转化生长因子 β1 ( TGF- β1 )对关节软骨细胞的影响。方法 :应用软骨细胞体外培养 ,分别加入 0 .1、1.0、10 .0 ng/ ml TGF-β1 ,用 EL ISA方法 ,测定培养液中的基质金属蛋白酶 3 ( MMP- 3 )浓度。结果 :TGF-β1 作用软骨细胞2 4小时 ,MMP- 3升高 ,但无显著性差异 ( P >0 .0 5 ) ;作用 48小时后 ,TGF- β1 使 MMP- 3降低 ,且与对照组比较有显著性差异 ( P <0 .0 1)。结论 :外源性 TGF-β1 具有抑制软骨细胞分泌金属蛋白酶的作用
【Abstract】 Objective: To explore the effects of TGF-β 1 on joint chondrocyte. Methods: Joint chondrocyte was cultured in vitro, added into TGF-β 1 of different concentration respectively. The matrix metalloproteinase-3 (MMP-3) levels in culture media were measured by ELISA. Results: After 24 hours treated, MMP-3 level raised, but it was no significant (P>0.05). After 48 hours treated, MMP-3 level fallen significantly (P<0.01). Conclusion: Exogenous TGF-β 1 inhibit the excretion of matrix metalloproteinase of rabbit joint chondrocyte.
