抗生素AGPM高产菌株选育及其蛋白差异分析

【摘要】 从土壤中分离得到一株能产生新型抗生素AGPM的藤黄灰链霉菌菌株.利用紫外线和硫酸二乙酯对新型抗生素AGPM产生菌藤黄灰链霉菌099菌株进行了联合诱变处理.最终得到了一株高产菌株TD0551,其抗生素AGPM产量可达到16.45mg/L,为出发菌株的2.1倍.应用蛋白质二维凝胶电泳技术分析了藤黄灰链霉菌的蛋白质组,比较了高产菌株与低产菌株在分泌AGPM过程中的蛋白质组差异.结果表明,在高产菌株的蛋白样品中有11个特异点,而在低产菌株的蛋白样品中有3个特异点,这些蛋白特异点可能与新型抗生素AGPM的合成有关.

【Abstract】 One strain of Streptomyces regensis was isolated from soil to produce a new antibiotic, and the original strains were mutated by the cotreatment of UV and diethyl sulfate. One high yield strain named TD0551 was screened to give a AGPM yield of 16.45 mg /L,which was 2.1 times higher than that of the original strain. The protein patterns of the culture of Streptomyces regensis were analyzed by using twodimensional polyacrylamide gel electrophoresis. Comparing with the original strain, eleven new protein spots were found and three protein spots were not found in the cultures of the improved strain. The results showed that the special protein might be related with the improvement of AGPM production from Streptomyces regensis.
