咯利普兰对NOD小鼠T淋巴细胞凋亡的影响

【摘要】 目的 通过对非肥胖型糖尿病 (NOD)小鼠T淋巴细胞凋亡率的分析 ,探讨咯利普兰 (rolipram)对 1型糖尿病的免疫调节机制。方法 将经环磷酰胺 (CYP)加速过的 60只体重相近的 4周龄雌性NOD小鼠随机分为两组 ,干预组给予咯利普兰 ( 8mg·kg-1) ,每日 2次 ,连续 30d ;对照组给予等次等量的磷酸盐缓冲溶液 (PBS)。将小鼠脾细胞制成单细胞悬液 ,通过DNA梯度图谱定性和流式细胞仪定量法分析外周T淋巴细胞的凋亡情况。结果 与PBS对照组比较 ,咯利普兰干预组发病率降低 (P <0 0 1) ;外周T淋巴细胞凋亡率增加 (P <0 0 5 )。结论 咯利普兰能够通过增加T淋巴细胞的凋亡减少免疫效应性细胞的积聚 ,从而阻止NOD小鼠发生 1型糖尿病

【Abstract】 Objective To investigate the mechanisms of immune regulation of rolipram by analysing the apoptosis of T lymphocytes in non-obese diabetic(NOD) mice.Methods Sixty female 4 week NOD mice (18 g) were randomly divided into two groups.Treatment group received the intraperitoneal injection of rolipram (8?mg·kg -1) twice a day.Control group received the intraperitoneal injection of PBS (equal quantity and times). The mice in two groups were administered cyclophosphamide(250?mg·kg -1) to accelerate the process of diabetes on day 1.All mice were killed on day 30 to observe the incidence of diabetes,the degree of insulitis and apoptosis rate of spleen lymphocytes were detected by means of DNA ladder qualitative analysis and flow cytometry quantitative analysis.Results Intraperitoneal injection of rolipram to NOD mice reduced the incidence of diabetes(P<0.01).The insulitis score and the severity of insulitis in rolipram group were lower than those in control group (P<0.01).The rate of apoptosis was higher in rolipram group (P<0.05).Conclusions Rolipram could prevent NOD mice from developing into type 1 diabetes by increasing apoptosis of T lymphocytes.
