类黄酮物质对染铂大鼠肾毒性防护作用研究

【摘要】 目的 :探讨类黄酮物质对顺铂 (CP)所致大鼠肾毒性的防护作用 ,为临床应用其防护顺铂肾毒性提供依据。方法 :以腹腔注射顺铂 5mg kg造成大鼠肾损伤模型 ,观察经口给予四种类黄酮物质后血尿素氮 (BUN)、肾皮质匀浆丙二醛 (MDA)和还原型谷胱甘肽 (GSH)含量的变化。结果 :提前给予四种类黄酮物质均可拮抗单次给予CP所致BUN、MDA升高和GSH的下降。结论 :类黄酮物质对顺铂致肾损伤有预防作用。其机理可能与类黄酮物质的抗氧化特性和抑制铂所致巯基耗竭有关

【Abstract】 Objective: To approach the preventive effects of flavones on nephrotoxicity of cisplatin(CP) in rats in order to provide basis for protecting nephrotoxicity of CP in clinic. Methods: Rats were given 5 mg/kg CP by peritoneal injection in order to induce nephrotoxicity .The levels of blood urea nitrogen (BUN), molondialdehyde (MDA) and glutathione (GSH) in renal cortex homogenate were compared between CP group and groups pretreated with four flavones orally. Results: The four flavones can resist CP-induced increase of BUN and MDA, and decrease of GSH in single dose. Conclusion: Flavones may protect kidney against CP-induced nephrotoxicity .The mechanism may be related to the effects that flavones against oxide injury and inhibit CP-induced sulfhydryl depletion.
