抗坏血酸抑制乙醇引起大鼠纹状体c-fos mRNA表达增强的多巴胺能机制
Dopaminergic mechanism of the inhibitory action of ascorbic acid on ethanol-induced c-fos mRNA expression in rat′s striatum
【摘要】 目的研究抗坏血酸对乙醇引起大鼠纹状体c fosmRNA表达增强的影响及与多巴胺能受体拮抗剂的相互作用。方法应用原位杂交法 ,观察纹状体内外两侧c fosmRNA的表达情况。结果一次性腹腔注射乙醇 ( 1 5、3 0 g/kg) 1h后 ,大鼠纹状体内外两侧c fosmRNA的表达均显著增强 ,外源性给予抗坏血酸 ( 10 0 0mg/kg)能显著抑制乙醇引起的表达增强。当多巴胺受体拮抗剂舒必利、SCH 2 3390、氟哌啶醇与抗坏血酸合用时 ,舒必利能协同抗坏血酸抑制乙醇引起的纹状体外侧c fosmRNA表达增强 ,而SCH 2 3390则协同显著增强乙醇引起的纹状体外侧c fosmRNA表达 ;氟哌啶醇协同抑制乙醇引起的纹状体内侧c fosmRNA表达增强。结论本研究在基因水平证实了抗坏血酸对乙醇引起的大鼠纹状体内外两侧c fosmRNA表达增强有显著的抑制作用 ,其抑制作用可能有多巴胺能神经系统参与调节
【Abstract】 Objective To study the effect of ascorbic acid on ethanol induced c fos mRNA expression in the rat′s striatum,and the relative dopaminergic mechanisms.Method To observe c fos mRNA expression in the medial and lateral striatum by in situ hybridization.Results Ethanol could induce c fos mRNA expression in rat′s medial and lateral striatum for 1 h after an acute administration of ethanol (1.5,3.0 g/kg,ip).Exogenous administration of ascorbic acid ( 1 000 mg/kg ,ip) significantly inhibited ethanol induced c fos mRNA expression. L sulpiride in combination with ascorbic acid synergistically was inhibited,but SCH 23390 and ascorbic acid potentiated ethanol induced c fos mRNA expression in lateral striatum.Haloperidol in combination with ascorbic acid synergistically inhibited ethanol induced c fos mRNA expression in medial striatum.Conclusion The studies show that ascorbic acid can significantly inhibit ethanol induced c fos mRNA expression in medial and lateral striatum of rats,and the dopaminergic neural system may be involved to mediate the inhibitive action of ascorbic acid.
