膀胱移行细胞癌中肿瘤细胞和间质细胞PD-ECGF的表达

【摘要】 目的 探讨血小板衍化内皮细胞生长因子 (PD -ECGF)在膀胱移行细胞癌肿瘤细胞和肿瘤旁间质细胞中表达的异同及其临床意义。方法 用PD -ECGF的单抗 1C6 - 2 0 3,采取免疫组织化学方法对 5 5例膀胱移行细胞癌和 8例正常膀胱粘膜进行PD -ECGF表达的研究。结果 正常膀胱粘膜的上皮细胞和间质细胞的PD -ECGF免疫染色均呈阴性。 5 5例膀胱移行细胞癌中 2 5例 (45 .5 % )肿瘤细胞PD -ECGF呈阳性表达 ,其肿瘤病理分级为G1级 2 / 7(2 8.6 % ) ,G2 级 9/ 2 5 (36 .0 % ) ,G3 级 14 / 2 3(6 0 .9% ) (x2 =3.91,P >0 .0 5 ) ;病理分期为Ta期3/ 18(16 .7% ) ,T1期 5 / 14 (35 .7% ) ,T2~ 4期 17/ 2 3(73.9% ) (x2 =14 .0 ,P <0 .0 0 1)。肿瘤间质细胞的PD -ECGF阳性表达为 37/ 5 5 (6 7.3% ) ,其病理分级为G1级 3/ 7(42 .9% ) ,G2 级 14 / 2 5 (5 6 .0 % ) ,G3 级 2 0 / 2 3(87.0 % ) (x2 =7.39,P <0 .0 5 ) ;病理分期为Ta期 5 / 18(2 7.8% ) ,T1期 12 / 14 (85 .7% ) ,T2~ 4期 2 0 / 2 3(87.0 % ) (x2 =30 .2 ,P <0 .0 0 1)。结论 膀胱癌肿瘤细胞与间质细胞中的PD -ECGF阳性表达在膀胱移行细胞癌的进展中都起着重要作用 ,检测其表达对判断膀胱癌的分化程度和浸润程度有一定临床意义

【Abstract】 Objective To analyze the expression difference of platelete-derived endothelial cell growth factor (PD-ECGF) between tumor cells and stroma cells in bladder transitional cell carcinoma(TCC) and its clinical implication. Methods Immunohistochemical staining with anti-PD-ECGF monoclonal antibody 1C6-203 was used to analyze the PD-ECGF expression of tumor and stroma cells in 55 cases of bladder TCC and 8 cases of nomal bladder mucosa. Results Normal bladder mucosa showed negative anti-PD-ECGF immuno-staining in both epithelial and stroma cells. Tumor cells expressed positive staining of PD-ECGF in 25/55(45.5%) cases of bladder TCC, Positive tumor cell PD-ECGF expression was found in 2/7(28.6%) cases of G 1,in 9/25(36.0%) cases of G 2 and in 14/23(60.9%) cases of G 3 tumors ( x 2=3.91, P >0.05);and was found in 3/18(16.7%) cases of Ta,in 5/14(35.7%) cases of T 1 and in 17/23(73.9%) cases of T 2~4 tumors ( x 2=14.0, P <0.001). Tumor stroma cells displayed positive PD-ECGF staining in 37/55 (67.3.%) cases of bladder TCC. Positive stroma cell PD-ECGF staining was found in 3/7(42.9%) cases of G 1,in 14/25 (56.0%) cases of G 2 and in 20/23(87.0%) cases of G 3 tumors ( x 2=7.39, P <0.05);and was found in 5/18(27.8%) cases of Ta,in 12/14(85.7%) cases of T 1 and in 20/23(87.0%) cases of T 2~4 tumors ( x 2=30.2, P <0.001). Conclusion Positive expression of PD-ECGF in both tumor and stroma cells might play an important role in the malignant progression of bladder TCC and may be used as a predicator of tumor dediferentiation and invasiveness in bladder cancer.
