新生大鼠脑缺氧缺血模型心肌细胞凋亡的研究
Apoptosis of myocardial cells in neonatal rat hypoxic-ischemic brain damage model
【摘要】 目的 研究新生大鼠缺氧缺血性脑损伤 (HIBD)发生后不同时段心肌细胞凋亡情况。方法 结扎新生 7日龄大鼠右颈总动脉后放入 8%低氧舱 2h制成HIBD模型 ,应用AnnexinV/PI双染流式细胞技术检测缺氧缺血 (HI)后 1、4、18、2 4、4 8、72h及 7d心肌细胞凋亡情况。结果 HI后各HI组心肌细胞的凋亡率均明显高于对照组 (P <0 .0 5 ) ,72hHI组凋亡率又显著高于其他组 (P <0 .0 5 ) ,表明HI后 7d内各组大鼠心肌均存在超出正常凋亡 ,尤以 72hHI组凋亡率最高。结论 新生大鼠HIBD后 1h~ 7d内心肌细胞均存在明显凋亡 ,以HI后 72h更明显 ,提示对缺氧缺血性脑病新生儿保护心肌减少心肌凋亡的治疗应至少用至生后 7d
【Abstract】 Objective To study apoptosis of myocardial cell after hypoxic ischemic brain damage(HIBD) in neonatal rat HIBD model.Methods The HIBD model was produced in the traditional model of neonatal hypoxia ischemia(HI) which subjected 7 old day Wistar rats to right carotid artery ligation followed by an hypoxic(8 % oxygen) episode of 2 hours duration. The proportion of apoptotic cells in myocardial cells was determined by Annexin V/PI double staining flow cytometry technique at time points of 1, 4, 18 , 24 , 48 , 72 hours, 7 days after HIBD.Results The average percentages of apoptotic myocardial cells in all HI groups were significantly higher than in the control group(P<0.05), and the average percentage in 72 h group was significantly higher than in other HI groups(P<0.05). It shows that myocardial cells of all rats in each HI groups had supernormal apoptosis, especially at 72 h after HIBD.Conclusions In neonatal rat HIBD model, there was significant apoptosis in myocardial cells during 1h to 7 days after HIBD, especially at the time point of 72 hours. It suggests that medication of protecting myocardial cells and reducing apoptosis of them for newborn with hypoxic ischemic encephalopathy should be taken for at least 7 days after birth.
