低分子量肝素的制备与纯化
Preparation and Purification of Low Molecular Weight Heparin
【摘要】 介绍以猪肠粘膜肝素粗品为原料,采用亚硝酸控制解聚法制备分子量在4000~8000D的低分子量肝素.采用正交实验法讨论反应温度、时间,亚硝酸浓度,溶液pH值等条件对解聚反应的影响.测定产物的分子量,经SephacrylS 200HR凝胶色谱柱对解聚的低分子量肝素进行分离纯化.由红外光谱13Cnmr谱图对其结构作鉴定.以粗品肝素作原料制备低分子量肝素,成本较为低廉,工业化前景比较广阔.
【Abstract】 A method of preparation of low molecular weight heparin from crude hog mucosal heparin by depolymerization with nitrous acid was studied. The optimization of the experimental conditions, such as temperature, time, concentration of nitrous acid and pH have been investigated. The depolymerized heparin having molecular weight of 4 000~8 000 Dalton was purified with column of Sephacryl S200. Average molecular weight of low molecular weight heparin were determined. The structure of the low molecular weight heparin was identified by using IR and 13CNMR spectra. Experimental results showed that method of preparation of low molecular weight heparin was satisfactory. The depolymerized product has lower cost from crude, So the technology of preparation of low molecular weight heparin from crude has broader prospect.
