【摘要】 目的　研究尿激酶型纤溶酶原激活剂 (uPA )在鼻咽癌侵袭和转移中的作用。方法　应用逆转录聚合酶反应 (RT -PCR )方法检测uPA在鼻咽癌细胞系 (CNE -2Z)及其克隆株 (CNE -2Z -H 5、CNE -2Z -H 5 -9)中的表达 ;应用酶联吸附法(ELISA )测定uPA在CNE -2Z、CNE -2Z -H 5、CNE -2Z -H 5 -9中的分泌量 ;采用异质黏附抑制实验 ,观察uPA在鼻咽癌细胞侵袭和转移过程中的作用。结果　uPA在CNE -2Z -H 5 -9中表达水平最高 ,在CNE -2Z中表达水平最低 ,在CNE -2Z -H 5中表达水平处于前两者之间 ;抗uPA抗体抑制CNE -2Z -H 5 -9的异质黏附能力。结论　uPA可能促进鼻咽癌细胞的侵袭和转移更多还原

【Abstract】 Objective To investigate the role of uPA in invasion and metastasis of NPC.Methods RT-PCR was used to detect the expression of uPA in NPC line(CNE-2Z) and its sub-lines(CNE-2Z-H5,CNE-2Z-H5-9).ELISA was used to detect the expression of uPA secreted by CNE-2Z,CNE-H5 and CNE-H5-9.The inhibiliting experiment of heterogeneous adhesion was used to observe the role of uPA in NPC′s metastasis and invasion.Results Expression of uPA was the highest in CNE-2Z-H5-9 and lowest in CNE-2Z.The expression of uPA in CNE-2Z-H5 was between that in CNE-2Z and CNE-2Z-H5-9.Anti-uPA antibody inhibited the heterogeneous adhesion of CNE-2Z-H5-9.Conclusion uPA may promote the metastasis and invasion of NPC.更多还原
