双染色-流式细胞术快速检测急性胰腺炎核因子-κB激活的研究

【摘要】 目的 建立特异敏感的快速检测急性胰腺炎核因子 κB激活的一种方法。方法  3 2只Wistar大鼠随机分为两组 :①胰腺炎组。采用 5 %牛磺胆酸钠逆行胰管内注射制备模型。②对照组。利用双染色 流式细胞术检测大鼠胰腺细胞NF κB的激活。结果 胰腺炎组各时段 (1 5、3、6、12h)NF κB和DNA双阳性细胞百分率明显高于正常对照组 ,P <0 0 1。 1 5hNF κB明显激活 ,为 (2 9 78± 7 83 ) % ,3h达高峰 ,其值为 (65 17± 13 2 2 ) %。 6h有所下降 ,但变化不显著 (P >0 0 5 ) ,12h明显下降 ,接近发病时水平。对照组各时段NF κB的活性变化不明显 (P >0 0 5 )。结论 流式细胞术是快速检测急性胰腺炎核因子NF κB激活的敏感特异方法

【Abstract】 Objective To develop a specific and sensitive way for rapidly detecting the nuclearfactor kappa B (NF κB) activation in acute pancreatitis (AP). Methods Wistar rats ( n =32) weredivided randomly into two groups.①Pancreatitis group:the acute panereatitismodel wasestablished by retrograde injection of 5% sodium taurocholate into the pancreatic duct.②Control group: double color flow cytometry was used to rats for detecting NF κBactivation in pancreatic acinar cells. Results It was found that the positive percentage of NF κB and DNA of AP group was significantly higher at each time point than that ofcontrol group( P <0 01). The NF κB activity [(29 80±7 83)%] significantly increased at h 1 5, with a peak[(65 17±13 22)%] at h 3. It decreased a little at h 6, and markedly decreased at h 12. The changes of NF κB activity of control group at different time points were not notable ( P >0 05). Conclusion Flow cytomety is aspecificand sensitive way for the rapid detection of NF κB activity in acute pancreatitis.
