1,25-(OH)2D3对NOD小鼠胰岛Bcl-2、Bax、TGF-β1、PCNA表达的影响
Immune mechanisms of 1,25-(OH)2D3 in preventing NOD mice from type 1 diabetes
【摘要】 目的 观察 1,2 5 (OH) 2 D3免疫调节作用与Bcl 2 ,Bax ,TGF β1,PCNA在胰岛的表达之间的相关性。方法 将 40只体重相近的 4周龄非肥胖糖尿病 (NOD)小鼠随机分为两组 ,每组 2 0只 ,干预组隔日腹腔注射 1,2 5 (OH) 2 D3(5 μg/kg) ,对照组注射等次等量的花生油。实验开始时 ,40只小鼠均一次性腹腔注射环磷酰胺 2 5 0mg/kg以加速胰岛炎。 3 0d处死 ,观察糖尿病的发病率、胰岛炎积分 ,Bcl 2 /Bax ,TGF β1,PCNA在胰岛的表达。结果 1,2 5 (OH) 2 D3处理组与对照组比较 :处理组发病率降低 (P <0 0 5 ) ,Bcl 2 /Bax比值升高 (P <0 0 5 ) ,TGF β1表达升高 (P <0 0 1) ;PCNA表达与 1,2 5 (OH) 2 D3无相关性。结论 1,2 5 (OH) 2 D3能够阻止NOD小鼠发生 1型糖尿病
【Abstract】 Objective To investigate the immune effect of 1,25 (OH) 2D 3 on diabetes in nonobese diabetic mice. Methods Forty 4 week female NOD mice were divided into 2 equal groups. Group 1 received theintraperitoneal injection of 1,25 (OH) 2D 3 (5 μg/kg) every other day. Group 2 received the intraperitoneal injection of peanut oil(equal quantity). Two groups’ mice were administered cyclophosphamide(250 mg/kg) to accelerate the process of diabetes. All mice were killed at day 30. When the experiment was finished,the incidence of diabetes was observed. The degree of insulitis and the expression of Bcl 2,Bax,TGF β1 and PCNA were detected with immunohistochemical technique. Results Intraperitoneal injection of 1,25 (OH) 2D 3 to NOD micereduced the incidence of diabetes(10% vs 55%, P <0 05).The ratio of Bcl 2/Bax was higher than that of control group〔(1 11±0 11) vs (0 37±0 16), P <0 05〕. The expression of TGF β 1 was higher than that of control group( P <0 01). The expression of PCNA in pancreatic islets is not correlated with 1,25 (OH) 2D 3. Conclusion These results show that 1,25 (OH) 2D 3 can prevent NOD mice from developing into type 1 diabetes.
