酶保护剂对提高酶稳定性的作用
The Function of Enzymatic Protective Reagent in Improving the Enzymes Stability
【摘要】 目的 观察酶保护剂对提高酶稳定性的作用。方法 研制出一种酶保护剂,并在酶溶液中加入酶保护剂,在一年中,对丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、乳酸脱氢酶(LDH)、碱性磷酸酶(ALP)和肌酸激酶(CK)的活性进行长期监测。结果 加入保护剂后,几种酶在室温下可以稳定2 w,在-10℃±5℃条件下可以保持稳定达一年。结论 酶保护剂可以提高ALT、AST、ALP、CK的稳定性和保存时间。保护剂对酶类的测定没有影响,所以加入酶保护剂的方法可以用来保存酶提取物和制备生化酶类质控物。
【Abstract】 Objective To observe the function of enzymatic protective reagent in improving the enzymes stability. Methods Developed an enzymatic protective reagent,and put it into enzymes solution. Determined the activities of alanine amino-transferase(ALT).aspartate aminotransferase (AST) ,lactate dehydrogenase(LDH) , alkaline phosphotase(ALP)and crea-tine kinase(CK) in the per od of one year. Results With this enzymatic protective reagent, enzymes were stable for 2 weeks in room temperature and 1 year in -10℃±5℃ condition. Conclusion Stabilities of ALT, AST, ALP and CK could be improved by using enzymatic protective reagent. There are no influences on enzymatic tests from enzymatic protective reagent. The method could be used in the preparation of enzymatic quality control serum and storing enzymatic isolate.
