携带猪β干扰素基因的鸡输卵管生物反应器逆转录病毒载体的构建
Construction of Recombinant Retroviral Vector Carrying Ovalbumin 5’-flanking Regulatory Sequence and Swine Interferon β Gene
【摘要】 为构建携带猪β干扰素基因的鸡输卵管生物反应器逆转录病毒载体 ,用PCR技术扩增了猪 β干扰素基因和卵清蛋白5’调控序列 ,然后将其重组到切除了Phsp70启动子的逆转录病毒载体pLNHX中。用酶切和PCR等方法对重组质粒进行鉴定 ,结果表明 ,卵清蛋白 5’调控序列和猪β干扰素基因已正确克隆至逆转录病毒载体pLNHX中。为进一步制备高滴度、高感染性的逆转录病毒 ,从而制作鸡的输卵管生物反应器奠定实验基础
【Abstract】 Swine Interferon β(IFN-β)and ovalbumin 5’-flanking regulatory region were amplifier by PCR Then they were inserted into vector pLNHX to generate a recombinant vector Pln-ov-in The recombinant vector were verified with restriction analysis and PCR The results showed that IFN-β and ovalbumin 5’-flanking regulatory region were cloned correctly into pLNHX It is concluded that the vector was constructed successfully for producing high-titer virus and making chicken oviduct bioreactor
