NASBA荧光分子信标技术定量检测丙型肝炎病毒
HCV Quantity Detection with NASBA Fluorescent Molecular Bacon Technique
【摘要】 建立NASBA荧光分子信标探针检测技术 ,并对国家HCV标准品、人工构建HCVRNA野生株及HCV抗体阳性不同人群进行检测。实验结果 :该方法检测HCV的灵敏度为 1 0 3 拷贝 ml血清 ,阴性参比品的符合率为 1 0 0 %;检测的线性范围为 1 0 3 拷贝~ 1 0 9拷贝 ml血清 ;精密性 (CV值 )小于 6%,在HCV抗体阳性人群中HCVRNA的检出率在 45 %~ 65 %之间。结论 :该方法在HCVRNA临床定量检测中具有良好的灵敏度、特异性、重复性与实用性。
【Abstract】 A NASBA fluorescent molecular bacon detection technique was established and was used in HCV quantity detection. The national HCV standard sample, wild type HCV RNA artifact and blood sera with HCV antibody positive from different people were detected respectively. Results: The HCV detection sensitivity in serum was 10 3 copies/ml using this technique; The consistency of negative control standard was 100%; The detective linear range was within 10 3 copies/ml to 10 9 copies/ml. The coefficient of variation (CV) was less than 6%. The positive rate of HCV RNA among HCV antibody positive people was within 45% to 65%. Conclusion: This RNA quantity technique is of good sensitivity, speciality, repetition and practicability in clinical HCV RNA quantity detection.
