利用β-环糊精键合固定相分离纯化葛根素

【摘要】 使用β 环糊精(β CD)与琼脂糖凝胶偶联合成的几种新型键合固定相对葛根黄酮进行液相色谱分离纯化,分离效果最好的介质为聚β CD偶联烯丙基 SepharoseHP所得的固定相,该固定相湿凝胶的样品负载量达1 21g/L。色谱分离时采用的流动相为10%(体积分数)醋酸水溶液,葛根素的收率在97%以上时,其纯度高于93%。

【Abstract】 Puerarin is a kind of Chinese traditional medicine, which has the good medical function to hypertension and angina. In this study puerarin was purified by lowpressure liquid chromatography. The experimental conditions, such as mobile phase, the stationary phase, loading capacity for the separation were optimized. Before separation, the Radix puerariae flavone was dissolved in 20% ethanol. The best stationary phase was polyβcyclodextrin coupled allylSepharose HP. The optimized separation condition was 10% (v/v) acetic acid as mobile phase with the flow rate of 1 mL/min. The loading capacity of the wet gel was 121 g/L for samples. Under these conditions, the purity of puerarin attained 93% with the recovery of 97%.
