反相高效液相色谱法测定全血中马钱子碱和士的宁的含量

【摘要】 建立了反相高效液相色谱法定量分析全血中马钱子碱和士的宁含量的方法。该方法应用OasisR ○MCX固相萃取小柱进行提取、采用XTerraTMRP18色谱柱进行分离。在该条件下马钱子碱和士的宁的线性范围为0 05~10mg/L,最小检出限为0 15ng。该方法测得马钱子碱和士的宁在低、中、高浓度下的回收率均在90%以上,日内、日间精密度均符合方法学要求。该方法灵敏、快速、准确,适用于临床血药浓度监测和法医毒化检验。

【Abstract】 A high performance liquid chromatographic (HPLC) method was developed for the si multaneous determination of brucine and strychnine in human blood. The samples w ere pretreated with solid phase extraction using Oasis R○MCX cartridges and the a lkaloids were separated on XTerraTM RP18 column with UV detection at 254 nm. Calibration curves were linear within the range of 005-10 mg/L an d the limit of detection was 015 ng for each alkaloid. The recoveries obtained from spiked blood for the t wo alkaloids were more than 90%. The intra-day and inter-day presisions were a ssessed at three mass concentration levels (05,10,20 mg/L), and the relati ve standard deviations were in the range of 17% to 94%. The method described was proved to be sensitive, rapid and accurate, and it can be applied to the id entification and determination of the alkaloids for forensic and therapeutic dru gs monitoring.
