茶叶指纹谱的毛细管胶束电动色谱法研究

【摘要】 用胶束电动色谱法建立了茶叶的色谱指纹谱并用于分析10种中国名茶。这种色谱指纹谱是由相对保留值(α)和相应的相对峰面积(Sr)组成的。该方法将色谱图转换成数据表,实现了图谱的数据化;并且以相对保留值表示各色谱峰的峰位,大大减少了各种操作条件对色谱相对保留值的影响,降低了色谱峰峰位变化的波动性,提高了样品间的可比性。这种色谱指纹谱在没有标准品的情况下也可得出可靠的、有意义的结果。优化后的色谱条件是:电泳电解质为硼砂0 02mol/L、十二烷基硫酸钠(SDS)浓度为0 025mol/L的溶液(pH7 0),分析电压20kV,紫外检测波长280nm,1 5kPa压力条件下进样10s,熔融石英毛细管内径为50μm、总长60cm、有效长度45cm。该方法不需要标样,操作简单、花费低,可为茶叶的质量评估提供科学依据。

【Abstract】 A fingerprint chromatogram of tea consisted of relative retention value (α) and corresponding relative peak area (Sr) was developed using micellar electrokinetic chromatography and applied to analyze ten well-known Chines e teas. The method converted chromatograms into data format and incorporated the relative retention values to show the peak identity, resulting in reducing the effects of operational factors and the fluctuations of peak retention and impro ving the sample comparability. The fingerprint chromatogram provided possibility to assess the quality of tea without standard samples. An electrolyte at pH 7 0 containing 0025 mol/L sodium dodecylsulfate and 002 mol/L borate was used. A potential of 20 kV was applied and the detection was performed by UV absorpti on at 280 nm. The tea samples were injected under 15 kPa pressure for 10 s. A fused-silica capillary with an internal diameter of 50 μm and a total length o f 60 cm was used. The effective separation length was 45 cm. The present met hod is simple, low cost and suitable for the quality assessment of tea.
