利用粟酒裂殖酵母生长法定量测定肌醇浓度

【摘要】 利用本实验室分离得到的一株天然的肌醇合成缺陷型的粟酒裂殖酵母 (Schizosaccha romycespombe)特异地依赖外部肌醇生长的特性 ,研究了该菌株在含有不同浓度肌醇的合成培养基中的生长情况 ,发现在一定的肌醇浓度范围内 ,菌株的生物量随肌醇浓度增大而增大 ,且呈线性关系 ,由此建立了一套微生物法定量测定肌醇浓度的方法。我们发现在固定的培养温度、振荡速度等外部条件下 ,当接种量OD60 0 为 1 0、葡萄糖浓度为 4 0 %时 ,在肌醇浓度 0~ 5mg/L范围内 ,培养 52h后 ,肌醇测定标准曲线最为理想 ,R值为 0 9973。应用该方法对标准肌醇样品的测定得到了较好的结果

【Abstract】 A strain of Schizosaccharomyces pombe with characteristic of the natural inositol auxotroph was used to study the growth in a complete synthetic medium with different concentration of inositol. It was shown that biomass of the strain increased with the concentration of inositol in specific concentration range of inositol and there was a linear relationship between them. So a microbiological method for quantitative determination of inositol was set up. It was found that the calibration curve was very satisfying (r=0.9973) when the inoculation size (OD 600 ) was 1 0,the concentration of glucose was 4.0% (w/v),the concentration range of inositol was from 0 to 5.0*#mg/L ,the cultural time was 52h and the other cultural conditions such as temperature and the shaking speed of the culture were fixed. The method for quantitative determination of inositol content was more simple and convenient than any other methods available.
