激光复合诱变选育木薯原料柠檬酸高产菌
Breeding of Citric Acid Producing Strains Using Cassava as Raw Material by Laser Compound Mutation
【摘要】 以柠檬酸生产菌黑曲霉 (Aspergillusniger) Wm -0 1为出发菌株 ,通过60 Co γ射线、硫酸二乙脂 (DES)及激光的复合诱变 ,采用高酸、高渗培养基筛选 ,从 2 46株突变株中筛选出一株木薯粉柠檬酸发酵高产菌株Wm 1 0 16,其发酵温度为 (3 9± 1)℃ ,周期 72h ,2 2 %木薯粉摇瓶 (平均总糖 17 9% ) ,产酸平均达 17 2 % ,15 0m3 大罐实验产酸 16 13 5 % ,周期 67h。
【Abstract】 Using Aspergillus niger Wm-01 as original strain,with the high acid contained and high osmotic pressure media as choosing media, a high citric acid producing strain,A niger Wml-016,from 246 mutants by 60 Co-γ ray,DES and laser compound mutation was selected In flask with the optimized fermentation temperature(39±1)℃and period(72 h),its average citric acid yield is 17 2% by using 22% cassava powder(average sugar content is 17 9%)as main raw material In industrial scale fermentor(150 m 3),its average citric acid yield is 16 135% while the fermentation period only 67 h
