内皮素受体阻滞剂对皮瓣坏死的影响

【摘要】 目的 研究内皮素受体A选择性阻滞剂BQ - 12 3对任意皮瓣的存活是否有影响。方法 在SD大鼠背部肉膜下形成任意皮瓣 ,分成用药组和对照组。测皮瓣存活率 ,取材进行HE染色光镜检查 ,使用激光多谱勒血流仪测量皮瓣血运情况 ,测SOD、GSH -PX的活性及MDA含量。结果 术后第 10天 ,用药组皮瓣的存活率显著高于对照组 (P <0 .0 1)。HE染色光镜检查见用药组的组织损害较对照组小。激光多谱勒测量发现用药组较对照组LDF值下降少 ,恢复快。用药组较对照组SOD及GSH -PX的活性下降少 ,恢复快 ,MDA值则上升少 ,恢复快。结论 内皮素受体阻滞剂BQ - 12 3可显著地提高任意皮瓣的存活率

【Abstract】 Objective To investigate whether ET receptor specific antagonist BQ-123 influences necrosis in random pattern skin flaps of rats. Methods 120 rats were designed randomly into 12 groups. We took two groups each time. BQ-123 were injected in group, the control group received an injection of drug-free normal saline. We measured surviving portion of the flap and observed the tissue by light microscope. Using Doppler ultrasound we measured the blood flow, MDA, SOD, GSH-PX were also measured. Results The flap survival area of BQ-123 injection group was significantly greater (P<0.01) than that of the control. Under light microscope, we found the tissue in BQ-123 group swelled less than the control. In every point (except 10 th day postoperatively and preoperation) the blood flow detected in BQ-123 group was significantly higher than that of the control group (P<0.01). In every point (except preoperation) SOD and GSH-PX in BQ-123 groups were significantly higher than those of the control groups (P<0.01). MDA were significantly lower than those of the control groups at 1 std, 4 thd, 7 thd postoperatively. Conclusion BQ-123, a kind of ET receptor specific antagonist, can significantly promote survival of skin flap.
