黄胸散白蚁纤维素酶的研究
Digestive cellulase from the damp-wood termite Reticulitermes flaviceps(Oshima)(Rhinotermitidae:Reticulitermes)
【摘要】 对黄胸散白蚁体内纤维素酶所做的研究结果表明 ,37℃时 ,C1酶与pNPC底物反应的最佳时间为 1h ,Cx 酶与CMC Na和 β 葡糖苷酶与Salicin反应的最佳时间均为 0 2 5h。黄胸散白蚁工蚁头部纤维素酶主要是Cx 酶和 β 葡糖苷酶。肠道组织中 ,3种纤维素酶主要分布于前肠和后肠组织。Cx 酶在前肠的酶量占肠道组织该酶总量的 5 2 9% ;C1和 β 葡糖苷酶绝大部分在后肠 ,在该部位所含酶量分别占整个肠道组织酶总量的 83 3%和 80 2 %。说明 3种纤维素酶的活性在不同组织中有较大差异
【Abstract】 The digestive cellulase from Reticulitermes flaviceps was determined in this study.The results indicated that when the cellobiohydrolase reacted with p-nitrophenyl-β-D-cellobioside at 37 ℃,the activity of enzyme reached the maximum at 1 hour after reaction.For endo-β-14-glucanase and β-glucosidase,the best reactive time was 0.25 hour when they reacted with CMC-Na and salicin separately.In the head of R.flaviceps (worker,)the endo-1,4-β-glucanase and β-glucosidase were main cellulases.As for the whole gut tissues,cellulases mainly distributed in the foregut and hindgut.Endo-β-14-glucanase in the foregut accounted for 52.9% of cellulases amount in the whole gut tissues.Cellobiohydrolase and β-glucosidase mostly located at the hindgut,at which they accounted for 83.3% and 80.2% of the total cellulases in gut tissue respectively.These results showed that the activities of cellulase varied in the different tissues in R. flaviceps.
