体外培养海马神经元NR1型受体表达的发育性变化
THE DEVELOPMENTAL EXPRESSION OF NR1 SUBUNIT ON CULTURED HIPPOCAMPAL NEURONS
【摘要】 用免疫细胞化学方法检测了体外培养的大鼠海马神经元不同培养时期的 N-甲基 -D-天门冬氨酸受体 1型亚单位的免疫活性反应强度 ,并用图象分析方法进行了定量研究。结果显示 :体外培养的海马神经元的 N-甲基 -D-天门冬氨酸受体 1型亚单位的免疫反应强度 ,在培养第 1d时较弱 ,第 2 d、第 4d和第 7d时反应强度逐渐加强 ,分别为 1d的 2 .44、3 .82和 4.18倍 ,随后反应强度稍有下降 ,第 10 d、第 14 d分别为第 1d的 3 .71和 3 .65倍。表明 :体外培养的海马神经元在发育进程中 N-甲基 -D-天门冬氨酸受体 1型亚单位表达进行性增加 ;当神经元发育成熟时 ,则其表达不再增加。提示 :N-甲基 -D-天门冬氨酸受体 1型亚单位与神经功能成熟有关
【Abstract】 The developmental expression of NR1 subunit of N methyl D aspartate receptor on in vitro cultured hippocampal neurons was demonstrated with the immunocytochemistry and the intensity of the immunoreactivity was measured quantitively with the image analysis system. The results showed that the immunoreactivity intensity of NR1 on in vitro cultured hippocampal neurons was weaker on 1 d,and progressively increased in 2.44 , 3.82 and 4.18 folds on 2 d, 4 d and 7 d, and then decreased slightly with 3.71 and 3.65 folds to 1d on 10 d and 14 d, indicating that with the culture ongoing in vitro development, the expression of NR1 increased progressively and became unchanged on matured neurons, suggesting that NR1 may be involved in the neural function maturation.
