乳头状甲状腺癌中转化生长因子-β及其受体的基因表达

【摘要】 目的 探讨转化生长因子 β(TGF β)及其受体基因的表达与乳头状甲状腺癌的关系。 方法 取甲状腺手术标本 ,其中乳头状甲状腺癌 8例、桥本甲状腺炎症 7例、甲状腺腺瘤 8例和正常甲状腺组织 8例 ,扩增TGF β1、TGF β2 、TGF β3 、TGF β受体Ⅰ、TGF β受体Ⅱ及TGF β受体Ⅲ基因 ,比较各种基因在不同组织中的表达水平。结果 TGF β1在 4种组织中表达阳性率均较高 ,但在乳头状甲状腺癌中的表达丰度高于其他 3组 (P<0 .0 5 )。TGF β受体Ⅰ在乳头状甲状腺癌组织中表达阳性率低于其他 3组 ,表达丰度也明显降低 (P <0 .0 5 )。TGF β2 、TGF β3 、TGF β受体Ⅱ及TGF β受体Ⅲ在 4种组织中均部分表达 ,且表达量的差异无显著性 (P >0 .0 5 )。结论 TGF β1表达水平上升和TGF β受体Ⅰ表达水平下降可能与乳头状甲状腺癌的发生有关 ,TGF β2 、TGF β3 、TGF β受体Ⅱ及TGF β受体Ⅲ基因与乳头状甲状腺癌可能不存在相关性

【Abstract】 Objective To study the relevance of gene expressions of transforming growth factor-β(TGF-β) and TGF-β receptor in specimens of papillary thyroid carcinoma(PTC), and their relationship with the genesis of PTC.Methods The thyroid tissue specimens of the patients were collected during the operation, including 8 papillary thyroid carcinomas, 7 Hashimoto thyroiditis, 8 thyroid adenomas and 8 normal thyroid tissues. The total RNA of all these specimens were extracted and purified by acidic phenol. The gene expression of TGF-β 1, TGF-β 2, TGF-β 3, type Ⅰ,Ⅱ, Ⅲ receptors of TGF-β(TGF-βR Ⅰ, Ⅱ, Ⅲ) were investigated by reverse transcriptional polymerase chain reaction(RT-PCR) and agarose gel electrophoresis. The degree of fluorescent electrophoretic bands were detected by automatic gel scanner and analyzed statistically to compare the levels of of the four tissue specimens.Results It was found that the gene expression of TGF-β 1 was high in all 4 groups, but the gene expression levels of TGF-β 1 in PTC was higher than that of the other 3 groups, showing presence of significant difference(P<0.05). In contrast, the positive rate of TGF-β R I in PTC was lower than that of the other 3 groups and the level of gene expression of TGF-βR I in PTC was also lower than that in the other 3 groups(P<0.05). The positive rate of TGF-βRⅡ gene expression was also high, but there were no significant differenas in the expression levels and positive rates between the four groups. TGF-β 2, TGF-β 3 and TGF-βRⅢ gene were expressed in a portion of specimens of the four groups, and no significant differences in the positive rates and the expression levels were found. Conclusion The increase of TGF-β 1 gene expression and decrease of TGF-βR Ⅰ gene expression may be related to the genesis of PTC, and can be used as a parameter for laboratory diagnosis of PTC. The expression of TGF-β 2, TGF-β 3 , TGF-βRⅡ and TGF-βRⅢ genes show no tissue specificity, therefore they seem to have no relationship between their expressions and PTC genesis.
