软枣猕猴桃超氧化物歧化酶的分离纯化与特性研究

【摘要】 目的从软枣猕猴桃果实中提取纯化超氧化物歧化酶 (SOD) ,并对其特性进行研究。方法先以硫酸铵分级沉淀 ,后经DE 5 2纤维素柱色谱 ,磷酸缓冲液进行梯度洗脱。结果得SOD干粉 0 .918mg ,其分子量约为 30 6 0 0D ,亚基分子量约为 15 2 0 0D ,最大紫外吸收波长为 2 5 4nm。结论软枣猕猴桃果实含丰富的SOD。

【Abstract】 PurposeA superoxide dismutase (Cu,Zn SOD) was extracted and purified from the fruit of Actinidia arguta (Sieb.et Zucc) Planth. Cu,Zn SOD and its properties were studied.MethodsCrude enzyme was obtained through (NH4) 2SO 4,and purified by DEAE Cellulose chromatography.Results0.918 mg of dry powder enzyme was obtained. This enzyme, has two identical subunits. The molecular weight is about 30 600,and the identical subunit molecular weight is about 15 200. The optimum temperature of this enzyme is at 25℃.The maximum absorbance of wave length is 254 nm.ConclusionThere is rich Cu,Zu SOD in the fruit of Actinidia arguta (Sieb.et Zucvc) Planth.
