骨髓基质细胞体外培养及成骨特性研究
Osteogenic Characteristic of the Cultured Bone Marrow Stromal Cells In Vitro
【摘要】 目的 建立兔骨髓基质细胞(BMSC)向成骨细胞转化的体外培养方法,并观察其体外的成骨特性。方法 利用静置贴壁原理进行BMSC的体外培养,汇合后传代,部分细胞改用条件培养液继续培养。对培养的细胞进行形态学观察和组织学检测,并观察条件培养液对BMSC的功能影响。结果 条件培养液组细胞碱性磷酸酶(ALP)染色阳性,细胞重叠生长形成多层结构,伴有散在的矿化结节形成。条件培养液组细胞ALP染色率及活性较完全培养液组高,但增殖率相对降低。结论 培养的BMSC经诱导具有体外成骨能力。地塞米松、β-甘油磷酸钠和维生素C可促使BMSC转化为成骨细胞,但抑制BMSC的增殖速度。
【Abstract】 Purpose To establish a culture method for bone marrow stromal cells (BMSC) differentiating to osteoblasts and describe osteogenic characteristic of BMSC in vitro. Methods Primary BMSC of rabbit were cultured and gradually purified from marrow cells suspensions due to their characteristic of adhering to plastics in vitro. Cells were subcultured at confluence, partly in a conditional medium containing dexametha-sone, β-glycerophosphate and ascorbic acid. Stromal cells were observed morphologically and histochemically. Effect of the conditional medium on BMSC were also examined. Results Histochemical examinations showed positive stain of alkaline phosphatase activity in BMSC of conditional medium group, in which a mul-ticellular layer and mineralized nodules were formed. Rate of positive satin and activity of ALP were en-hanced,.while proliferation of celis were inhibited by treatment with conditional medium. Conclusions Af-ter proper induction,cultured BMSC can form bone tissue in vitro. This may provide a rich cellular resource for bone tissue engineering.
