【摘要】 目的　观察β 榄香烯对人胃癌细胞系SGC - 790 1凋亡抑制蛋白survivin及caspase酶活性的影响 ,探讨β 榄香烯诱导SGC - 790 1凋亡的作用机制。 方法　用不同浓度 β 榄香烯作用胃癌SGC - 790 1细胞后 ,分别采用Northernblot、Westernblot检测SurvivinmRNA和蛋白水平 ,用酶标仪比色法检测Caspase酶活性 ,分别采用TUNEL法检测细胞凋亡。结果　β 榄香烯能下调SurvivinmRNA和蛋白水平 ,增强Caspase酶活性 ,并诱导胃癌SGC - 790 1细胞凋亡 ,效果呈浓度和作用时间依赖性。结论　β 榄香烯能诱导人胃癌细胞系SGC - 790 1凋亡 ,可能与下调Survivin表达与增强Caspase酶活性有关。更多还原

【Abstract】 Purpose To study the expression of Survivn and Caspase activity of human gastric carcinoma cell line SGC-7901 treated with β-elemene,and to investigate the mechanism of β-elemene to induce tumor cell’s apoptosis. Methods After gastric cancer SGC-7901 cell were treated with different doses β-elemene,the protein and mRNA level of Survivn were detected by Western blot as well as Northern blot respectively,Caspase activity were evaluated by microplate reader,apoptosis were studied by TUNEL. Results β-elemene could down-regulate the level of protein and mRNA of Survivn,and elevate the Caspase activity,and induce gastric cancer SGC-7901 cell apoptosis.The effects were be relevant to the time and dose of β-elemene. Conclusions β-elemene could induce SGC-7901 cells apoptosis by down-regulation of expression of Survivin and up-regulation of Caspase activity.更多还原
