虫草多糖对大鼠肝星状细胞核因子-κB活性和肿瘤坏死因子-α表达的影响

【摘要】 目的 探讨虫草多糖对大鼠肝星状细胞的作用机制。方法 分离大鼠肝星状细胞 ,采用不同浓度的虫草多糖处理后 ,分别应用MTT法、凝胶迁移率法、ELISA法和Northernblot检测大鼠肝星状细胞的增殖、核因子κB(NF κB)活性和肿瘤坏死因子 α蛋白和mRNA的表达。结果 虫草多糖对大鼠肝星状细胞生长具有较强的抑制作用。虫草多糖可明显抑制大鼠肝星状细胞核因子κB活性和下调肿瘤坏死因子 α蛋白和mRNA的表达 ,并且呈剂量依赖性。结论 虫草多糖对大鼠HSC增殖具有很强的抑制作用 ,可能与抑制NF κB活性和下调肿瘤坏死因子 α表达有关

【Abstract】 Purpose To study the mechanism of cordycep polysaccharide on hepatic stellate cells. Methods After hepatic stellate cells of rat were treated with cordycep polysaccharide,proliferation,activity of nuclear factor κB,expression of protein and mRNA of tumor necrosis factor-α were studied by use of MTT,electorphoretic mobility shift assay,enzyme-linked immunoadsordent assay(ELISA) and Northern blot. Results Cordycep polysaccharide had a remarkable supression effect on hepatic stellate cells of rat,and cordycep polysaccharide repress the activity of nuclear factor κB,and down-regulate expression of cytokine tumor necrosis factor,which were dose-dependent. Conclusions It is suggested that cordycep polysaccharide repress the proliferation of hepatic stellate cells through inhibition of the activity of NF?κB and the expression of TNF?α and lowering the level of protein.
