灰飞虱β3-微管蛋白cDNA的克隆及序列分析

【摘要】 抽提灰飞虱RNA,利用RT-PCR技术,克隆了灰飞虱体内β3-微管蛋白基因部分序列。进一步通过3’-和5’-RACE获得了全长。序列分析表明,该基因(LSTUB3)长1859bp,编码的蛋白含454个氨基酸,分子量约为51kDa;类似于其他昆虫的β-微管蛋白,LSTUB3含有2个氨基酸保守区:NNWAKGHY和RKAFLHWYTGEGMDEMEFTEF。另外,LSTUB3还具有1个由6个氨基酸组成的β3-微管蛋白特征性的插入序列:CASRGG。结合已发表的其他昆虫该基因的序列,构建了分子系统树,系统分析表明:LSTUB3与其他昆虫的该基因序列有较高的同源性,达85.7%以上。

【Abstract】 The mRNA isolated from adult Laodelphax striatellus was reverse transcribed. The cDNA was PCR amplified using degenerate primers designed according to the alignment of the β 3- tubulin amino acid sequences of other species. To complete the coding sequence, the 3’ and 5’ rapid amplification of cDNA ends (RACE) was performed. The full- length cDNA of β 3- tubulin gene of L. striatellus (LSTUB3) spans 1859 bp and encodes a protein of 454 amino acid. The amino acids sequence predicted for the putative protein is highly homologous to β 3- tubulin characterized from various insects. It shares two conserved amino acid sequence motifs NNWAKGHY and RKAFLHWYTGEGMDEMEFTEF with other insect tubulins. In addition, there is an insertion of a six amino acid sequence from LSTUB3 residue 56- 61, which is a significant characteristic of β 3- tubulin.
