牛未成熟卵母细胞体外受精及染色体分析

【摘要】 收集不同供体牛的卵母细胞并分成两组。体外成熟培养16h为不完全成熟组;体外成熟培养24h为完全成熟组。结果显示,体外受精48h后,成熟组大部分胚胎发育到8细胞期和16细胞期,不完全成熟组的多数胚胎发育到2细胞期和4细胞期,染色体异常发生率显著高于成熟组。染色体异常表现为单倍体、多倍体和混合倍体。引起染色体异常发生的原因可能是由于精子穿入时透明带或卵母细胞膜的功能障碍所引起。

【Abstract】 Bovine oocytes collected from different donors were pooled into two groups. Oocytes matured in vitro for 14 h were designated as the insufficient maturation group, and other oocytes matured in vitro for 24 h as the maturation group. At 48 h after insemination, most of the embryos in the 24 h group developed into the 8 or 16 cell stages, while many embryos in the 16 h group were at the 2 or 4 cell stage. The incidence of chromosomally abnormal embryos in the 16 h group was significantly higher than that in the 24 h group. These chromosomal anomalies were haploids, polyploids and mixoploids. It was presumed that the increase in these abnomalies was caused by sperm penetration with dysfunction of zonae pellucida and/or membrane of the immature oocytes.
