5-氟脲嘧啶诱导人胆囊癌细胞凋亡的研究
The effect of fluorouracil on apoptosis in humangallbladder carcinoma cell
【摘要】 为研究 5 -氟脲嘧啶 (5 - FU)对胆囊癌细胞凋亡的影响 ,在体外培养的胆囊癌细胞中加入不同浓度的 5 -FU ,应用光镜、电镜、DNA凝胶电泳及流式细胞仪检测癌细胞凋亡的形态学特征、生化学特征、细胞凋亡百分率及细胞周期变化。在 5 - FU作用下 ,凋亡的胆囊癌细胞出现膜小泡、凋亡小体等特征性改变 ;DNA电泳呈现典型的“梯状”条带 ;流式细胞仪测定出现典型的凋亡峰 ;其凋亡百分率随着药物浓度的提高而明显升高 (与对照组比较 ,P<0 .0 5 )。认为 5 - FU可诱导胆囊癌细胞发生凋亡 ,其凋亡率随药物浓度的提高而升高
【Abstract】 To study the effect of Fluorouracil on apoptosis in human gallbladder carcinoma cell line GBC-SD. Human cultured gallbladder carcinoma cell GBC-SD was exposed in vitro to different concentrations of Fluorouracil for various length of time. Cell apoptosis was ascertained by cell morphology under light-, and electron microscope,and DNA electrophoresis and flow cytometry.When GBC-SD cells were treated with Fluorouracil at different concentration, cell apoptosis occurred. Typical apoptotic bodies appeared and DNA ladder could be demonstrated on DNA electrophoresis. Apoptotic peak was also shown by flow cytometry. The percentage of cell apoptosis increased with raised concentrations of drug, and compared with the control group cells, the difference is significant(P<0.05). Fluorouracil successfully induced the apoptosis of human gallbladder carcinoma cells, and the percentage of cell apoptosis increased with raised concentrations of drug.
