9-顺-维甲酸对人肺鳞癌细胞周期调控因子CyclinD1、Cdk4转录的影响
Effects of9-cis-retinoic acid on CyclinD1and Cdk4transcription in lung cancer cell line
【摘要】 目的:检测9-顺-维甲酸(9-cis-RA)处理前后人肺鳞癌细胞株L78、A2的细胞周期调控因子Cy-clinD1、Cdk4转录水平的变化。探讨其抑制肺鳞癌细胞生长的作用机制。方法:采用半定量逆转录聚合酶链式反应(RT-PCR)分析细胞周期调控因子CyclinD1、Cdk4转录水平的变化。结果:9-cis-RA对两细胞株的细胞周期调控因子CyclinD1、Cdk4转录水平均有所降低,且具有统计学意义(P<0.05)。结论:9-cis-RA抑制肺鳞癌细胞株L78、A2增殖的作用机理与其调节细胞周期调控因子CyclinD1、Cdk4的转录有关。
【Abstract】 Objective:To study the changes of CyclinD1and Cdk4mRNA level in human lung cancer cell line L78and A2before and after treatment with9-cis-RA(1μmol/L),and to explore the ef-fecting mechanism of9-cis-RA on the proliferation of lung cancer cells.Methods:The mRNA expressions of CyclinD1and Cdk4of human lung cell line L78and A2were determined by RT-PCR.Results:9-cis-RA significantly reduced the mRNA expressions of Cdk4and CyclinD1in lung cancer cell line L78and A2.Conclusion:The growth inhibitory effect of9-cis-RA on lung cancer cells is tightly correlated with its declining the mRNA expression of Cdk4and CyclinD1.
