利用细菌血红蛋白提高大肠杆菌基因工程菌几丁质酶基因的表达
Vitreoscilla Hemoglobin Enhances Chitinase Production and Secretion of a Recombinant Escherichia coli Strain
【摘要】 将透明颤菌(Vitreoscilla)血红蛋白基因(vgb)插入几丁质酶基因表达质粒中,然后转入大肠杆菌(Escherichiacoli)内,研究vgb基因的表达对几丁质酶基因表达和产物分泌的影响.结果表明,在不同的通氧条件下,vgb基因的表达产物均能不同程度地促进细胞的生长,而且发酵上清液的几丁质酶活性也有提高;若溶氧浓度越低,则效果越明显,可比对照提高达12.1%.同时,由于vgb在大肠杆菌的表达,还可进一步促进几丁质酶在大肠杆菌中的分泌.
【Abstract】 A recombinant plasmid pCV23 was constructed by inserting the Vitreoscilla Hemoglobin (VHb) gene into the plasmid pCHI, which contained the chitinase gene cloned from Bacillus circulans C2. To study the influence of VHb to cell growth and chitinase production, both plasmids were transformed into Escherichia coli JM109, respectively. The results showed that the expression of VHb could improve cell growth in different dissolved oxygen concentrations. And the production of chitinase was enhanced with 12.1% higher than that of control. It was observed that VHb also influenced the chitinase secretion.
