消痰散结方对人胃癌细胞周期及CDK4表达影响的实验研究

【摘要】 [目的]观察消痰散结方(主要由半夏、胆南星、鸡内金、全蝎等组成)对胃癌细胞周期及CDK4表达的影响,以探讨其抑制胃癌增殖的机理。[方法]将人胃癌MKN-45接种于裸鼠腋部皮下,分别给予小鼠消痰散结方、环磷酰胺或生理盐水21 d后处死小鼠,取出瘤组织,用流式细胞仪检测各组胃癌组织中处于G1、S期的细胞数,用免疫组化SABC法检测胃癌组织中CDK4的表达。[结果]中药组G1期细胞数为(66.84±1.14)%,空白组为(54.66±8.72)%,二者比较有显著性差异(P<0.01),中药组、化疗组S期细胞比例均明显低于空白组(P<0.01),中药组S期细胞数也明显低于化疗组(P<0.01)。中药组胃癌组织中CDK4表达总阳性率、中度及以上阳性率均明显低于空白组和化疗组(均P<0.05)。[结论]消痰散结方可明显抑制胃癌细胞的增殖;其机理可能是通过抑制CDK4的表达来影响细胞周期的完成。

【Abstract】 [Objective] To observe the effect of Xiaotan Sanjie Prescription (XSP) on human gastric carcinoma cell (GCC) cycle and CDK4 expression and to explore its mechanism in inhibiting the proliferation of gastric cancer. [Methods] Nude mice were inoculated with human gastric carcinomatous tissue (MKN-45) into subaxillary region subcutaneously and then were treated with XSP (Group A), cyclophosphamide (Group B) and normal saline (Group C) respectively. Twenty-one days later, the mice were executed and the tumor tissues were obtained. G1- and S- phase GCC count in carcinomatous tissue was carried out by flow cytometry and CDK4 expression by immunohistochemical method. [Results] G1-phase GCC accounted for (66.84±1.14)% in Group A and (54.66±8.72)% in Group C, the difference being significant ( P < 0.01). The number of S-phase GCC in Group A and Group B were lower than that in Group C ( P < 0.01), and that of Group A lower than Group B ( P < 0.01). Positive rate of total CDK4 expression and that of middling positive expression and above in Group A were both lower than those in Group B and Group C (P < 0.05). [Conclusion] XSP can inhibit the proliferation of GCC and its mechanism may be related to the interference of cell cycle by inhibiting CDK4 expression.
